In the absense of Resident Evil 6 or any JRPG, I'll go with Halo Anniversary. Skyrim would have got it, but everything they showed had been revealed prior.

Zelda: Skyward Sword. I was very meh on the game prior to E3, now I really want to play it.
Paper Mario 3DS. I love these games so much. The interplay between 2D and 3D gameplay makes the game so much fun to play. Now that the 3D will be even better I can't help feeling very excited about it

From Dust, Insanely Twisted Shadow Planet, Journey and Bastion were shown. They are all going to be BIG!
Besides the big names (like ME3), I would have to say I am most excited for
- Dragon's Dogma (PSO meets Grffins meets Monster Hunter but mostly action? Yespleasethanks!)
- Dragon's Crown (anything Vanilla Ware!)
- The Lord of the Rings: War in the North (SNOWBLIND!!)
- Kingdoms of Amalur (I was pimping this game ealier in the year and no one took notice!)
- BioShock Infinite (Sorry!)
Can't decide on GOTS 'cuz I really haven't seen much of anything FROM the show besides quick snippets and descriptions from quick hands-on previews!
Skyword Sword.
It's the only thing I've watched more than 30 seconds of. I've watched about a minute of footage. 
Can I just be a dick and say nothing? There just wasn't much new stuff shown this year, as far as the games I was interested in.
I guess maybe Bioshock Infinite. I watched more footage of that than I had before and am now a little more interested in it.
I have seen hours and hours and hours of E3 stuff but I still have more to see.
But game of the show for me is Zelda, its been decided already.
Second place right now is Skyrim.
Zelda, definetly Zelda. It looks gorgous, they fixed all the things that were wrong with the older Zelda, it has awesome motion+ combat and puzzles and dungeons in dungeons in dungeons. Plus flying and shit.
Zelda looks incredible,as well as Uncharted 3. It's a toss up between these two. At the moment, I'm more hyped for Uncharted but I'm sure that's because of all the lovin' I've been giving the second one at the moment. I just know my GOTY will be a toss up between those two.
