If we are only talking about this gen, then I'd pick the Wii. There are very few exclusive games on PS3/360 that really stand out to me, and scream, "Must play!" The vast majority of the games released on those platforms just don't appeal to me.
I'd go with the 360 myself.

Even though all I have is the Wii, it's exclusives are pretty much the reason why I got it in the first place and don't really have any serious desire for a second console. It's where most of the good niche games are:

Rune Factory: Frontier
NyxQuest
Bit.Trip Beat
De Blob
Muramasa
We Ski & Snowboard
MadWorld
And of course all the 1st party exclusives from Nintendo are icing on the cake.
There's still a few on the PS3 I'd love to get my hands on. Really would like to have my own copy of FLOWer.
About PS3 vs Wii, sorry its no contest, my Wii doesn't get close the attention that the PS3 does. Of course this is because of 3rd party as well. As for exclusive versus exclusive, I still have to go with PS3. Sure Nintendo may have three of the greatest franchises of all time but there is a big drop off after that. While with the PS3 I get exclusives in almost every genre and many in my favorite, action/adventure.
Ok list time, exclusives for the PS3 that make the system:
MGS4 - best game this gen
Little Big Planet - other best game this gen, except that it isnt just a game, its the most unique game I have played this gen, incredible.
Uncharted 1 and 2 - Part 1 was a great game, the second one takes it to a whole different level and it is easily one of the best games this gen.
Ratchet ToD and ACIT - Best platformers not named Mario, though they are almost more action games than platformer.
Infamous - Awesome sandbox game, one of the best.
God of War 3 - Again one of the very best action/adventure games this gen.
Demon's Souls - the most interesting RPG I have played in years, the secret is that it plays nothing like an RPG, it plays like an action game.
Valkyria Chronicles - Only played the demo so far but it was awesome, will play when I get around to it.
Motorstorm 2 - Fun arcade racing.
Heavy Rain - Again yet to play but its very unique
Killzone 2 - Good single player campaign and a great online mode.
PixelJunk Eden - I love this game, weirdo scoring platformer thing
Wipeout HD - Its wipeout so its fun
Soon we get Gran Turismo 5 and Last Guardian.
And Wii of course has a ton of exclusives due to the fact that basically every game made for it has to be different than the other two. Which is why I feel you can get by this gen with a Wii and one of the HD systems cause the two HDs share so many of the same games. Wii has one library, HD has another one. Anyway here are some of my favorite Wii exclusives

Wish I could get pumped to make a decent response. But I'm kinda the guy who thinks that all systems -after a reasonable period of time- have enough great exclusives to justify owning.
I prefer wii exclusives because I get the High Quality first party games and then really different, interesting and high quality stuff like Endless Ocean, Silent Hill, Red Steel 2 etc.
I would take a Zack and Wiki over your everyday blockbuster sequel any day.
To me, PS360 are interchangeable. Sure, it's nice to delve on the exclusives each system offers, which is the reason I picked them both up, but if pressed, I'd be OK with having only one of them.
Wii is nice to have as an extra, but I couldn't live on my Wii alone. I have it precisely for the games that other systems don't offer, games that throw you for a loop with originality, but my tastes are located too far into the mainstream territory to be content with Wii titles alone, and besides, 1st party Nintendo efforts mean little to me (mainly the holy trinity of Mario/Zelda/Metroid).

I'm like that in many ways also. I'd prefer games like A Boy and His Blob or Muramasa over any exclusive the PS3 or 360 offers...but I do admit I can't live on that alone.
I do need some blockbusters like Bad Company 2, New Super Mario Bros. Wii, or Uncharted 2 (when I get a PS3 someday) for my gaming needs.
But I would purchase A Boy and His Blob over Uncharted 2 if I had to get one...even though I will get Uncharted 2 later on, for example.

Well, I guess I was just posting mostly Wii's first-party Ninty games, but yeah, you're right, although I listed Tatsunoko VS Capcom and Monster Hunter Tri on there.
And I disagree about KZ2 or RE2. RE2 maybe, but KZ2 is I think one of the best FPSs this gen.

Yeah. But I will get it this year. I just need some more time. Hope Angel is having fun with his PS3!
Also, like I mentioned, I don't know about R2 but I think KZ2 is definitely great. I had a chance to play it this past week and I thought it was pretty awesome, aside from the motion blurring that hurt my eyes.
What other FPSs are there that could give it a run for its money? All I can think of is Halo, Modern Warfare 2, Bad Company 2, and all the other FPSs. KZ2 is certainly better than most of the smaller FPS titles, and I think the single player is probably better than the other big guns.
Multiplayer is another story, of course. I still think Bad Company 2 is the best multiplayer experience of this gen so far.
As for competing against the 360 exclusives...I agree with you there. Although I'm not a Mass Effect fan and I haven't played ME2 yet, so I can't say much for that. So I'll just take peoples' word on it if they claim it's better than KZ2. But I will say that Gears of War 2 is arguably a lot better than KZ2, but it's all about taste I guess. That's one example.

And just these titles alone is better than my 40 360 games. And if you count multi-platform games, the PS3 is the better system in many ways.

In many ways, I'd rather have these than any of the 360 exclusives or games. Of course, being a Nintendo fan helps.We always talk about the Wii being a great companion console to have. Having the Wii by itself isn't enough to get the full experience, but as I think about this, I think the PSWii combo is superior to the Wii360 combo.
Of course it's all about taste. But I think what I'm trying to say is that the PS3 is the best system IMO. The first-party exclusives have been the best so far. I think the Wii is a close second because Nintendo is still the best developers in the world.
I still love my 360 but it does have its shortcomings. The 360's saving grace is the online play and that multi-platform games usually tend to be better for some bizarre reason. But just thinking about Uncharted 2 and Killzone 2 makes me jealous, that's all I have to say.
