To answer your questions, you can play games with the 360 controller or wathever other controller you want. I even saw mod that made it possible to play Half-Life 2 with a Wiimote and chuck.

It depends on the genre of the game, really. Like Iga said, RTS games, turn-based strategy ones and most modern WRPGs (Mass Effect, The Witcher, Dragon Age) have interfaces that take full advantage of the KB/M combo, so playing them with a controller is not only pointless but also disadvantageous. I am pretty sure most PC games have the option of playing with a controller, like Assassin's Creed, GTA and so on.
